Comparison of Postmortem Neuropathology in E280A PS1 Mutation Carriers with
Sporadic Late-Onset Alzheimer's Disease

   Proposal Description:
   The goal is to compare comorbid neuropathology in the brains of 100 E280AD PS1  mutation carriers with that in sporadic late-onset AD subjects with data at       NACC.  The hypothesis is that comorbid pathology, if prevalent in PS1 mutation   carriers, is mechanistically linked to AD molecular pathogenesis rather than an  independent, age-dependent change. Comorbid pathology types to be investigated   are those recorded in the NACC NP10 data form, and include Lewy body             (alpha-synuclein) pathology, TDP-43 pathology, gross and microscopic infarcts,   and non-AD tauopathies including argyrophilic grains and progressive             supranuclear palsy. The 100 PS1 mutation cases are derived from a source         separate from NACC but will be analyzed with neuropathology protocols             equivalent to the 2012 NIA-AA guidelines (Hyman et al, 2012 Montine et al         2012), using the NACC NP10 form.
